NMR relaxometry as a potential non-invasive routine sensor for characterization of phenotype in Crassostrea gigas

چکیده
12 MR imaging is the most appropriate non-invasive technique for quantifying the growth of 13 somatic and gonad tissues and to determine sex in the Pacific oyster, Crassostrea gigas. 14 However, this technique is too costly for field studies where oysters are used as bioindicators of 15 environmental quality or to be applied routinely in hatcheries. We have tested the ability of low 16 Nuclear Magnetic Resonance relaxometry, a much less expensive technique, to obtain phenotype 17 parameters that can be used to monitor the physiological state of oysters. NMR measurements 18 were carried out at three different periods using a low field spectrometer equipped with a 50 mm 19 diameter probe to investigate 60 oysters in their first year of maturity, which were then dissected 20 to measure internal shell cavity volume and dry flesh weight and to determine sex and gonad 21 development. The NMR results showed that it was possible to determine both internal shell 22 cavity volume and dry flesh weight in less than one minute with very high determination R 23 coefficients (0.95 and 0.94, respectively). The results showed also that it was possible to identify 24 sex and gonad development, with success rates of 93% and 83%, respectively. For oysters with 25 dry weight above 0.7g, the success rate in identifying sex was 100%. متن کاملExploitable Lipids and Fatty Acids in the Invasive Oyster Crassostrea gigas on the French Atlantic Coast
Economic exploitation is one means to offset the cost of controlling invasive species, such as the introduced Pacific oyster (Crassostrea gigas Thunberg) on the French Atlantic coast. Total lipid and phospholipid (PL) fatty acids (FAs) and sterols were examined in an invasive population of C. gigas in Bourgneuf Bay, France, over four successive seasons, with a view to identify possible sources ...
متن کاملThe density and spatial arrangement of the invasive oyster Crassostrea gigas determines its impact on settlement of native oyster larvae
Understanding how the density and spatial arrangement of invaders is critical to developing management strategies of pest species. The Pacific oyster, Crassostrea gigas, has been translocated around the world for aquaculture and in many instances has established wild populations. Relative to other species of bivalve, it displays rapid suspension feeding, which may cause mortality of pelagic inv...
متن کاملIn vivo RNA interference in oyster--vasa silencing inhibits germ cell development.
This study investigated the potential of RNA interference, which is technically challenging in bivalve mollusc species, to assess gene function in the oyster Crassostrea gigas. We designed dsRNA targeting the oyster vasa-like gene (Oyvlg), specifically expressed in oyster germ cells. In vivo injection of oyvl-dsRNA into the gonad provokes a knockdown phenotype corresponding to germ cell underpr...
متن کاملNMR assignment of the apo-form of a Desulfovibrio gigas protein containing a novel Mo-Cu cluster.
We report the 98% assignment of the apo-form of an orange protein, containing a novel Mo-Cu cluster isolated from Desulfovibrio gigas. This protein presents a region where backbone amide protons exchange fast with bulk solvent becoming undetectable. These residues were assigned using 13C-detection experiments.
